Protective Effect of PRP against Testicular Oxidative Stress on D-galactose Induced Male Rats

Abstract

D-galactose (d-gal) administration known to increase production of Reactive Oxygen Species (ROS) and decrease antioxidant enzymes, causing oxidative stress in various organs, such as the testes. Oxidative stress triggers lipid peroxidation process which is characterized by the production of Malondialdehyde (MDA) products. The activation of Necrosis Factor Kappa Beta (NFkß) due to oxidative stress causes an increase in the production of pro-inflammatory cytokines, such as Interleukin-6 (IL-6). Moreover, increase levels of MDA and IL-6 in the testes cause testicular damage. Growth factors in platelet-rich plasma (PRP) play a major role in tissue regeneration, having biological effects as anti-inflammatory agents and antioxidants. The benefits of PRP in male reproduction need to be developed further. This study aimed to determine the effect of PRP on testicular oxidative stress of Wistar rats induced by d-gal. Rats were induced by d-gal (300 mg/kg body weight) with daily intraperitoneal injections and intratesticular PRP injections administered once a week for 6 weeks. The statistical analysis showed significantly lower levels of MDA and IL-6 on testicular of rats group induced by d-gal with PRP compared with d-gal without PRP group (p<0.001). Conclusively, injection PRP intratesticular can reduce MDA and IL-6 levels in testes of rats.
