Abstract
Potato dextrose agar (PDA) is typically used as a medium for cultivating fungi. However, because fungal culture media can be expensive, it is essential to explore more affordable, organic alternatives to traditional media for microbial studies. This study assessed the effectiveness of alternative media-based rice molasses agar (RMA), corn molasses agar (CMA), barley molasses agar (BMA), and algae molasses agar (AMA) in supporting fungal growth compared to standard PDA. All media sustained fungal development, although growth rates and colony morphology varied. Fusarium exhibited significantly slower growth in media lacking a carbon source, underscoring the importance of carbohydrates for fungal proliferation. Chemical analysis showed that maize had the highest carbohydrate content, while barley had the highest protein content. Fusarium, Pyrenophora, Aspergillus, and Trichoderma isolated from wheat and barley plants or seeds responded differently across media. Barley and algae media supported growth comparable to or greater than PDA, with algae notably enhancing germination. Rice flour performed the weakest overall. These findings highlight the potential of barley and algae media as cost-effective, nutrient-rich alternatives to commercial fungal media, offering reduced contamination risk and broad applicability for fungal cultivation.
