Genetic diversity of nectar yeast from central Java based on RAPD method

Abstract

Plants use nectars to attract pollinators, so the chemical composition of nectar affects the pollinator and plant interaction. Yeast and other microorganism have been discovered to grow inside the nectar. The goal of this research was to determine the diversity of yeast in the nectars from flowers of Baturraden Botanical Garden and Panggeran flower nursery using Random Amplification of Polymorphic DNA (RAPD). The nectars were inoculated in Tauge Extract Agar. The DNA from pure yeast isolates was extracted using Chelex 100® Resin (Bio-Rad Laboratories, Inc.) and amplified using polymerase chain reaction (PCR) applying published M13 and CDU primers. The PCR products were analyzed using electrophoresis. The similarity of isolates was determined using Jaccard’s coefficient from the band patterns. Clustering was done using the UPGMA method. The result shows that all yeast isolates were grouped into two clusters; yeasts from Baturraden botanical garden belongs in one cluster, while the yeast isolates from Panggeran flower nursery spread in both clusters. Two isolates are not part of both clusters. Disagreement of this study to other finding on morphological, biochemical and physiological characters needs to be elucidated further.
