Abstract

The purpose of this study is to determine the regeneration power of soybean embryo explants callus at various concentrations of 2,4 D growth hormones and BAP in vitro. This study was conducted at the Laboratory of Bio-Science and Plant Reproduction Biotechnology, Department of Agricultural Cultivation, Faculty of Agriculture, Hasanuddin University (UNHAS), Makassar, from December 2019 to February 2020. Data were analyzed using analysis of variance, BNJ 5%. The results showed that the growth regulator 2,4 D (auxin) had no significant effect on the time and the percentage of callus formation, but had a significant effect on the callus weight and callus color, which resulted in a concentration of 2.4 D between 8 ppm - 10 ppm and better callus color. BAP growth agent (cytokinin) singly had no significant effect on the time and proportion of soybean embryo callus explants, but giving 1.0 ppm of BAP concentration had a significant effect and could improve callus color quality, while 2.0 ppm of BAP could increase callus weight. The combination of growth regulators 2.4 D 10 ppm and BAP 2 ppm produced better callus wet weight, that is 3.35 grams.
