Experimental studies of Alzheimer’s disease have largely\rdepended on transgenic mice overexpressing amyloid precursor\rprotein (APP). These mice, however, suffer from artificial\rphenotypes because, in addition to amyloid b peptide (Ab),\rthey overproduce other APP fragments. We generated knock-in\rmice that harbor Swedish and Beyreuther/Iberian mutations\rwith and without the Arctic mutation in the APP gene.\rThe mice showed typical Ab pathology, neuroinflammation\rand memory impairment in an age-dependent manner.
