Immune-Pineal Axis - the idea is that the production of melatonin no only shuttles between central (pineal) and peripheral (immunocompetent cells) sources, but is function changes from transducer of darkness (chronobiological function) to a defense molecule (inhibitor of cell adhesiveness, anti-oxidant).
First question--- which of the function was the primitive in the evolutionary scale?

Circulating melatonin primes endothelial cells in a long-lasting manner. The expression of adhesion molecules, such as PECAM and ICAM-1 on the plasma membrane in cultured endothelial cells reflects the circulating concentration of melatonin at the hour that the rats were killed (Tamura et al., 2010).
This paper increases the importance of the Immune-pineal axis in controlling the state of activation of endothelial cells.
We do not know whether this could be extended to other cells and its importance in choosing cells for cell therapy

Elaine - primeira pergunta - quem é o estressor - a própria luz ou a falta de melatonina?
Poderia ser melatonina vinda da pineal ou de outra fonte?
Tem já vários trabalhos mostrando que a ingesta de triptofano induz a produção de melatonina que tem ação local, mas pode ir à circulação.

Eu lembro bem do teu trabalho da melatonina como antidepressivo.

Este modelo que você mandou na semana passada parece bem interessante.
Caso houvesse uma redução, ou mesmo recuperação da depressão induzida pela luz com melatonina isto poderia indicar que a luz não seria a causa, mas sim a supressão noturna de melatonina.

Mas aí vou viajar.... Afinal estivemos em Copen no verão... imagine no inverno... altamente depressivo. Portanto, ratos mantidos em claro - condição em que gostariam de estar nos braços de morfeu, faria com que eles ficassem deprimidos.

Em outro paper mostramos que melatonina, em doses de suplemento, age como antidepressivo em modelo de UCMS (umpredictable chronic mild stress) em camundongos. Como o modelo de depressão deste paper usa a luz como stressor principal (e o modelo parece bem bom para induzir correlatos de depressão em camundongos) a idéia é explorar como investigar melatonina aqui. Como suplemento e ver se tb funciona, mas tb ver como mexer com tudo isso através de picos de luz ou sei lá o que. Cronobiológos que se manifestem.

About this group

The pineal gland produces melatonin at night in a daily basis. During the mounting of an innate-immune response nocturnal melatonin is suppressed, allowing a proper migration of leukocytes to the site of lesion. Lately, activated immune-competent cells synthesize melatonin, which will act as an anti-oxidant, phagocytic and anti-inflammatory mediator.
