This chapter discusses conjugational methods for mapping with Hfr and F-prime strains. Conjugational mapping methods are particularly useful when the map location of a locus is totally unknown, and there is no clone available for molecular mapping of the locus, or when a clone bearing an antibiotic resistance marker within or near an unmapped locus has been inserted in the chromosome by homologous recombination. Hfr strains with other points of origin have also been isolated. It is also possible to tailor conjugal donor strains using small F or F-prime (F') factors which carry some homology to a site which has been located on the chromosome. The chromosome mobilization from such a strain results in oriented transfer of donor markers beginning at the site of homology between the F derivative and the chromosome. © 1991, Elsevier Inc.
