● Granulocyte macrophage-colony stimulating factor (GM-CSF) can be applied in treatment of the neonatal hypothyroidism and neuroendocrine dysfunction. Thus, neonatal GM-CSF may be responsible for suppressing autoimmune responses. ● The maternofetal thyrotoxicosis by Levo-thyroxine (L-T4) appears to alter the developing HPTA, GH/IGF and cytokines/adipokines axes. The maternal treatment with Complete Freund’s Adjuvant (CFA) seems to reduce or prevent the fetal neuroendocrine dysfunction via regulating HPTA and monoamines. Thus, the maternal protection by CFA can be applied in treatment of maternofetal thyrotoxicosis and fetal neuroendocrine disorders. More interestingly, this protection is dependent on dose, duration, developmental period, and the species involved, as well as the exact mechanism of this improvement is still unclear. ● Normal maternal THs may be required for healthy offspring. ● The endocrine disrupting compounds can produce complex, mosaic effects during the life cycle. Conclusively, loss of body weight may suggest a decline in the general health level of animals, which can be important in the interpretation of thyroid effects. ● Exogenous thyroid hormone inhibits IGF-1-stimulated glucose uptake and cell proliferation in L6 myoblasts. ● Fetal/neonatal thyroid dysfunction may be related to the tested doses (Weaning PCB 95, Maternal TCDD or MMI, Lactating PTU and Gestational DOX). ● Thyroid transporters (THTs) and deiodinases (Ds) are expressed in brain in a region- and cell-specific way. ● THTs and Ds are important regulators of intracellular T3 availability and therefore contribute to the control of thyroid receptors (TRs)-dependent CNS development and early embryonic life. ● Defects in fetal THTs (MCT8 and OATP) or Ds may have more impact on fetal brain since they can result in intracellular T3 deficiency despite sufficient maternal TH supply. These deficiencies can adversely affect neurodevelopment but the neurological phenotypes are far from understood. ● Any small changes in thyroid function during the development may result in brain damage. ● Generally, the developing thyroid-brain axis may be very sensitive to any disturbance during the gestation/lactation periods. ● Disruption of the developmental neuroendocrine system related to the tested drugs/compounds may limit their efficiency in human life. ● The moderate iodine deficiency causes an imbalance in maternal thyroid homeostasis, especially toward the end of pregnancy, leading to isolated hypothyroxinemia suggestive of biochemical hypothyroidism. ● Uncontrolled hypothyroidism in pregnancy can lead to preterm birth, low birth weight and mental retardation. ● The disturbance in the maternofetal THs due to antithyroid drugs, Levo-thyroxine or environmental disruptors may lead, in turn, to the biochemical dysfunctions, morphofunctional alterations, developmental abnormalities and brain damage if not corrected prior/after the birth. ●The long-term effects of gestational diabetes may influence, generally, on the health of the embryos and newborns. Also, it may produce a developmental delay, embryopathy, geno-and cyto-toxicity, teratogenic changes, free radicals, and apoptosis. ● The obesity of mothers and disturbance in adipokines prior to pregnancy are a significant risk factor for the disturbed development of the pregnancy and the child, and develop metabolic syndrome. Thus, maintaining normoglycaemia and adipokines levels during pregnancy may play an important role in a healthy life for the newborns. ●The oral administration of Gonadotropin-releasing hormone (GnRH; LHRH- A2) may induce ovarian cycle and play vital gonadotrope role during the folliculogenesis process in ducks. ● However, it remains to be clarified whether the reported data in animal models may be significant to human health.
