Abstract

Abstrak. Streptozotocin (STZ) dapat menginduksi terjadinya diabetes melalui kerusakan sel beta pankreas. Penelitian sebelumnya menyatakan bahwa pemberian STZ dosis rendah secara bertahap dapat menimbulkan perbaikan spontan kadar gula darah pada tikus diabetes. Penelitian ini bertujuan untuk mengamati perubahan kadar glukosa darah puasa (GDP) setelah pemberian tunggal STZ dosis rendah (35 mg/Kg i.p) selama 12 minggu. Enam belas ekor tikus putih jantan Spraguen-Dawley dengan rerata berat badan 214,40 ± 8,25 g, dibagi dua kelompok yaitu kelompok kontrol dan tikus yang diinduksi STZ dosis rendah. Tikus kontrol hanya diberikan larutan NaCl fisiologis secara intraperitoneal. Kadar GDP diukur setiap 2 minggu selama 12 minggu pasca induksi STZ dan dibandingkan kedua kelompok. Pada akhir minggu ke-12 kadar GDP masih menunjukkan hiperglikemia (172,77 ± 14,57) dan berbeda bermakna dibandingkan kadar GDP hari ke-3 pasca induksi (p = 0,011). Data kedua kelompok pada akhir minggu ke-12 pasca induksi juga memperlihatkan perbedaan bermakna (p = 0,004) yang menunjukkan belum terjadi perbaikan spontan kadar GDP. Pemberian tunggal STZ dosis rendah (35 mg/Kg i.p) dapat meningkatkan kadar GDP dan belum terjadi perbaikan spontan kadar GDP yang diamati setelah 12 minggu pasca induksi (p = 0,004). In a previous study was conducted that multiple low doses of streptozotocin (STZ) can induce spontaneous restoration of normoglycemia in diabetic rats. This study was designed to investigate the fasting blood glucose (FBG) disturbances after the low dose streptozotocin administration (35 mg/kg i.p) for 12 weeks. Sixteen male Sprague-Dawly rats weighing 214,40 ± 8,25 g were divided into two groups, control and single low dose streptozotocin induced diabetic rats (35 mg/Kg i.p). Control rats were given only physiological saline i.p. Fasting blood glucose level was measured every 2 weeks for 12 weeks after induction and compared to other groups. At the end of twelfth week, FBG levels still showed the hyperglycemia (172.77 ± 14.57) and was different significantly than third day post-induction (p = 0.011). FBG levels of both groups was also different significantly (p = 0.004) which indicated no spontaneous restoration. Study found that single low dose streptozotocin (35 mg/Kg i.p) may increase of fasting blood glucose and no spontaneous restoration of normoglycemia were observed after 12 weeks (p = 0,004).
