Resistance to multiple anti-cancer drugs can arise de novo or as a result of chemotherapy of cancer. Expression of the drug efflux pump, P-glycoproten (MDR1), is a contributing cause of failure of chemotherapy. Understanding of the normal function of P-glycoprotein in handling drugs in the body, and its role in conferring anti-cancer drug resistance, has led to new strategies to treat cancer and deliver drugs throughout the body.
