The aim of this research was to develop and test a RP-HPLC/ELSD analytical procedure which can be both a suitable method for milk triacylglycerol determination and a useful tool for pre-separation of TAG fractions for further GC/MS identification. This application can be usefully adopted when a Liquid Chromatography/Mass Spectrometry apparatus is not available. RP-HPLC technique, with the conditions chosen, provided, in less than 20 minutes of analysis, a TAG profile including 30 peaks, most of them well resolved. A simple, home-made Stream Splitter System (SSSy) was assembled and used to collect, before the ELSD entrance, both milk fat triacylglycerol fractions and pure triacylglycerol molecules, which were then submitted to GC/MS analysis for identification. Finally the RPHPLC/ELSD technique was applied for the evaluation of the adulteration of milk fat with different amounts (5-50%) of beef tallow and palm kernel oil. The preliminary results suggested that even amounts of the adulterating fats lower than those tested can be detected by the data obtained by RP-HPLC/ELSD profiles.
