Type 2 diabetes (T2D) and obesity are epidemic problems worldwide. These disorders are linked to decreased life expectancy and increased incidence and mortality from cancer. Understanding the mechanisms of cancer progression in obesity and T2D will aid in discovering cancer prevention and treatment strategies for these patients. Hyperinsulinemia, insulin receptor, and insulin-like growth factor-1 (IGF-1) receptor signaling appear to be important factors linking diabetes, obesity, and cancer. A number of mouse models have been developed in order to accurately understand the roles of insulin, IGF-1, and their receptor signaling in obesity and diabetes-related cancer. This chapter summarizes many of the mouse models used to study T2D and cancer.
