Abstract

In this present study, two extraction methods of chlorogetic acid in honeysuckle were investigated including pectinase extraction method (PEM) and ultrasonic extraction method (UEM). It aimed to study factors influencing each extraction efficiency of chlorogetic acid by designing single factor experiments. Also, It aimed to investigate optimal conditions of each extraction method by visual analysis method and variance analysis method based on L9(43) orthogonal experiment and choose a relatively efficient extraction method by using extraction rate of chlorogetic acid as the index. The chlorogenic acid yield of using PEM and UEM were higher than traditional water extraction and ethanol refluxing method. The optimal extraction conditions in pectinase method were obtained as follows: solid- liquid ratio 1:30 (g/mL), the temperature of treatment 50 °C, pH = 5, dosage of pectinase 0.5%, treatment time 120 min, the yield of chlorogenic acid was up to 4.75%. The optimal extraction conditions in ultrasonic extraction were treatment temperature 45 °C, ultrasonic power 280 W, ultrasonic time 80 min, ethanol concentration 70%, solid-liquid ratio 1:30 (g/mL), the yield of chlorogenic acid was up to 6.26%.
