Abstract

Background: The Sec61 lateral gate mediates transmembrane segment insertion into the ER membrane. Results: Interactions and the effects of mutations of the Sec61 lateral gating helix residues were determined. Conclusion: Key residues that modulate translocation, membrane insertion, and topogenesis of membrane proteins are identified. Significance: This study offers mechanistic and structural insights into Sec61 gating. © 2014 by The American Society for Biochemistry and Molecular Biology, Inc. Published in the U.S.A.
