Abstract

Mouse transgenesis by pronuclear injection generates random integration events resulting in variable overexpression efficiency. This chapter will outline current strategies for improved transgene design (using plasmids or bacterial artificial chromosomes) and delivery to the mouse genome. The choice of genetic background of embryos for microinjection will also be discussed. © 2009 Humana Press, a part of Springer Science+Business Media, LLC.
