A short overview on principles and applications of fluorescence measurements is given, and three methods are described in detail. These methods include total internal reflection fluorescence (TIRF) microscopy and screening, fluorescence anisotropy as well as Förster resonance energy transfer (FRET) and total internal reflection energy transfer (TIRET) microscopy. All these methods have a high potential in biomedical research and in vitro diagnostics, but also need further optimization and standardization. The present chapter shows some approaches towards those goals and describes possible applications.
