Bio-molecular structure and dynamics have been studied using different fluorescence monitoring methods. In the present review, we have discuss the principle and application of different methods used to monitor conformation and dynamics of proteins (beta-lactamase, barnase, Ribonuclease) and DNA (duplex, triplex and quadruple), and macromolecules assembly (membrane) etc. We have also discussed the rules that are used to monitor the conformation of biomolecules using intrinsic and extrinsic fluorescence as well as selective monitoring of surface tryptophan of protein (beta-lactamase) using excitation at 305 nm. Therefore, the present fluorescence studies will be useful to monitor the bio-molecular structure and dynamics.
