Ca 2+-sensitive electrode as a practical approach is used to follow Ca 2+ changes in the medium and particularly useful to study mitochondrial Ca 2+ uptake (or release); this method permits the continuous recording of Ca 2+ movements through the mitochondrial inner membrane. In this chapter, it is described how to prepare a Ca 2+-sensitive electrode, and its application on mitochondrial studies with emphasis on the mitochondrial permeability transition. © 2012 Springer Science+Business Media, LLC.
