Abstract

SynopsisAs part of a large multi-protein molecular machine (the replisome), the helicase engine and the primase interact structurally and functionally with each other and with several other replisomal proteins. One of the most important interactions is that between the helicases and their loader proteins. But since this topic has been discussed elsewhere (initiation of replication), it will not be included in this section.IntroductionReplicative helicases are the power engines driving large replisomal assemblies. They form structural parts of replisomes and through their intimate interactions with various replisomal components and associated accessory factors directly modulate the function of the replisome in a variety of ways. The Helicase-Clamp-Loader InteractionThe bacterial replicative helicase is tethered onto the lagging strand and translocates unidirectionally in the 5′-3′ direction driving the replication fork forward. Its association with the replisome is mediated via a direct ...
