Abstract

A perfusion culture with an inclined cell settler for cell retention was used to produce human growth hormone. Cell densities of 9.0106 cells/mL could be maintained in steady state over a designated period of time to increase the process productivity. The cell extraction and perfusion rates had to be finely adjusted to maintain steady state conditions. Analyses of growth hormone concentrations were performed by ELISA and RP-HPLC. The downstream procedure comprises several techniques including centrifugation, ultradiafiltration and hydrophobic interaction chromatography.
