Advances in biomedical imaging are now impacting the evaluation of gonads and gametes in experimental animal model systems and human clinical specimens. This chapter summarizes the rationale for both live cell and fi xed sample imaging using light microscopy. Useful protocols, detailed methods, and tools for data extraction are covered for a variety of applications pertaining to the analysis of oocytes, embryos, and intact ovarian tissue.
