OBJECTIVE: To analyze the levels of Neu5Ac by instrumental methods. METHODS: High-performance capillary electrophoresis was applied to separate the glycoforms derived from different levels of Neu5Ac in rHuEPO. RESULTS: The phosphate buffer of pH 4.0 could deactivate the capillary wall effectively, so that the best resolution was obtained because of the reduced electroosmotic flow. CONCLUSIONS: The HPCE method established here could be employed to effectively detect the purity, microheterogeneity and in vivo bioactivity.
