Abstract

PCR amplification of Mortierella alpina cDNA using primers corresponding to conserved histidine-rich regions of known desaturases resulted isolation of cDNA clone Ma29, which showed significant homology to the known DELTA6-desaturase from cyanobacteria. Two additional cDNA clones were identified by random sequencing; Ma648 with homology to plant microsomal DELTA12 desaturases and Ma524 with homology to borage DELTA6 desaturase. All three desaturases were expressed in rape and changes in fatty acid composition of transgenic seeds were monitored.
