Tofu waste is the waste that comes from making tofu. Tofu waste is used by some people as food for livestock. The high protein and carbohydrate content of tofu waste is used as an alternative medium to grow mushrooms. One of the fungi used is the genus Penicillium sp.This study aims to determine the composition of the best tofu waste flour media for the growth of Penicillium sp. This type of research is an experimental laboratory with independent variables of dextrose sugar mass variation (0.2.4 grams), while the dependent diameter (cm) of Penicillium sp growth and macroscopic morphological characteristics are macroscopic. The dependent variables observed were compared with Sabouraud Dextrose Agar (SDA) media as a standard gold media. The composition used in this study was per 100 ml of distilled water consisting of 5 grams of tofu waste flour, 2 grams of pepton, agar 1.5 grams and dextrose sugar (0.2.4 grams). Based on the results of the study using statistical test the morphological growth characteristics of Penicillium sp using Kruskal wallis showed 0.983 results meaning there was no significant difference while the results of the statistical growth test of Penicillium sp diameter using Anova One Way showed 0.020 results meaning that there were significant differences. It can be concluded that Penicillium sp grows on alternative media of tofu waste flour. The best growth with the addition of 2 grams dextrose mass variation.
