Difference two-dimensional-infrared (2D-IR) spectroscopy has been employed to measure the 2D-IR spectrum of the C=C and C=ND chromophore bands in bacteriorhodopsin, demonstrating that couplings between individual bands in a mid-size protein can be measured.
