Abstract

Transmembrane (TM) proteins fulfil many crucial cellular functions and make up a large fraction of any given proteome with estimates suggesting up to 30% of all human genes may encode alpha-helical TM proteins. However, relatively few high resolution TM protein structures are known, making it all the more important to extract as much structural information as possible from amino acid sequences. In this chapter, we discuss current topology and structure prediction methods against a background of knowledge that has been gleaned from membrane protein sequence and structures analysis. We attempt to highlight potential pitfalls and identify major issues yet to be resolved. © 2009 Springer Netherlands.
