An endo-β-1,3-glucanase with a molecular mass of 33.2 kDa was purified from Mitsuaria chitosanitabida H12 for the first time. The optimal temperature and pH were at 52°C and pH 5.2, respectively, with laminaran as the substrate. The K m and V max were 1.145 mg/ml and 0.634 μmol min -1 ml -1, respectively, with the initial velocity solved by hyperbolic equation fitting and seeking the first-order derivative at its origin (0) point. After optimization, 20 and 50 mg/ml, 1,3-β-D-glucooligosaccharides of different lengths could be produced from yeast β-glucan with the enzyme. © Springer-Verlag and the University of Milan 2011.
