Abstract

Declining in potato harvested area and conventional systems that are still widely used in black potato cultivation demand an alternative way of production, one of which is through in vitro. This study aims to find out the response of black potato plants in various acclimatization media by applying golden snail liquid organic fertilizer. This research was conducted at the Tissue Culture Laboratory and Horticulture Development Garden State of Polytechnic Jember, Rembangan, Jember Regency in August-November 2022. The experiment was arranged using a Factorial Complete Randomized Design with 2 factors and 4 replications. The first factor was the type of acclimatization media for husk charcoal and fern root with ratios of 1:3, 1:1, and 3:1. While the second factor was the concentration of golden snail liquid organic fertilizer of 20 ml/L, 50 ml/L, and 100 ml/L. The results showed that there was an interaction between the type of acclimatization media and the concentration of gold snail LOF on the root length (49.42 cm). Husk charcoal media and fern root (3:1) was the media with the highest root length (46.68 cm). The concentration of gold snail LOF  of 20 ml/L was the best concentration to be applied to black potatoes acclimatized with plant height (54.18 cm), number of branches (70.08), internode length (15.16 cm), and stover weight (202.78 g)
