Abstract

To study the effect of astragalus polysaccharide on function of erythrocyte in tumor model mice. S180 mice were made as tumor model mice and treated with APS by i.p. for 7 days, and then the erythrocytes were collected. Band 3 and GPA content were determined by SDS-PAGE, test kits were used to measure phospholipids, cholesterol and sialic acid contents, DPH dye was used to determine membrane fluidity. Erythrocytes were labeled by rat anti-mouse CR1 antibody and FITC-conjugated goat anti-mouse antibody, flow cytometry was used to determine the number of CR1, then the DTER and RBC-C3bRR was determined by microscope. The results showed that Astragalus polysaccharide increased phospholipids content and decreased cholesterol content of erythrocyte membrane in tumor model mice, and increased both Band 3 and GPA contents and sialic acid content, also it improved erythrocyte membrane fluidity. Asparagus polysaccharide even increased CR1 number and activity of erythrocyte in tumor model mice. These results indicate Astragalus polysaccharide can adjust the abnormality of erythrocyte and improve its function. © Springer Science+Business Media Dordrecht 2014.
