Raman spectroscopy can be used to identify and analyze molecules in a sample without any labeling or modification. Since Raman scattering spectra provide information about intracellular molecular distributions, metabolism, or chemical reactions, Raman microscopy is widely utilized for bio-imaging and bio-functional analysis. By using metallic nanoparticles, Raman scattering from molecules at the vicinity of the metal surface is significantly enhanced. Due to localized field enhancement, highly sensitive detection of molecules can be achieved. In this review, we introduce a microscopic technique using Raman scattering for the observation of molecular dynamics in living biological cells. We also discuss the use of metallic nanoparticles for enhancement of Raman scattering from intracellular molecules for the functional analysis of various biological events.
