Abstract

The following steps for intiating, selecting, maintaining, and regenerating plants from type II callus and type II callus-derived suspension cell cultures represent a general format that has proven successful for inbred A188 and A188 derivatives (Armstrong and Green 1985; Kamo and Hodges 1986). The following protocols and the timing of specific steps should be used primarily as a guideline; they will require fine-tuning for different maize lines. In addition, we stress that visual selection and testing for plant regeneration from the type II callus and suspension cells must be done continuously.
