The application of fluorescence cross-correlation spectroscopy (FCCS) to measure molecular binding is examined. Technical challenges and solutions are discussed. Comparisons are made between one-photon excitation, two-color FCCS and two-photon excitation FCCS are made. Applications of FCCS to ligandreceptor binding and to lipid membrane fusion are presented.
