Abstract

Since the first single-molecule fluorescence resonance energy transfer (FRET) measurement in 1996, the technique has contributedsubstantially to our understanding of biological molecules and processes by probing the structure and dynamics of nucleicacids, protein molecules, and their complexes with other molecules. This review discusses basic concepts and current developmentsin single-molecule FRET methodology, as well as examples of applications to systems such as nucleic acid machines and molecularmotors.
