A review on the recent progress in the design and development of prodrugs of phosphonate, phosphinate, and phosphate functional groups to improve their physicochem. properties, membrane permeability, oral bioavailability, and drug targeting. Topics discussed include key issues of designing prodrugs for mols. carrying a phosphonate, phosphinate, or phosphate group, assay systems to evaluate the phosphonate, phosphinate, or phosphate prodrugs, and their specific examples.
