Altered cellular energetics associated with mitochondrial dysfunction is implicated in the etiology of multiple neurodegenerative diseases emphasizing a strong need for reliable and reproducible methods of detection. The Extracellular Flux (XF) Analyzer from Seahorse Bioscience provides an outstanding opportunity for comprehensive assessment of mitochondrial function and cellular energetics by monitoring real-time aerobic respiration and glycolysis in intact, living cells. In this chapter, we provide a method to measure respiration in primary mouse cortical neurons using a Seahorse XF24 Analyzer.
