Abstract

Human cancer results from the accumulation of multiple independent genetic alterations. Studies of inherited and sporadic colorectal cancer have demonstrated that in the overwhelming number of cases the primary muta- tion targets a single signal transduction pathway.Astory has rapidly unfolded in which the Wnt signaling pathway plays a central role in the etiology of this cancer. The sequence of discoveries provides a classic example of the synergy between insights gained from genetic stud- ies of model organisms and cancer biology in man.
