The discovery of a small-molecule allosteric inhibitor of the CDC34 ubiquitin-conjugating enzyme (E2) by Ceccarelli et al. raises the possibility that it will be generally feasible to selectively inhibit ubiquitin transfer at this central step in the ubiquitin pathway. © 2011 Elsevier Inc.
