Abstract

Processes are provided for formation of colloidal systems of nanoparticles of a substance (proteins, lipids, pharmaceuticals, etc.). In the case of proteins (for particle size <500 nm), the process comprises combining (1) a 1st liq. phase consisting essentially of the protein in water or in an aq. mixt., the 1st liq. phase being at a temp. lower than than the coagulation temp. of the protein; and (2) a greater vol. of a 2nd liq. phase consisting essentially of water or an aq. mixt. at a temp. higher than the coagulation temp. of the protein, which may contain a biol. active substance and to which may be added ≥1 surfactants; thereby substantially instantaneously to ppt. from the 1st liq. phase and the 2nd liq. phase particles of the protein to produce a colloidal suspension of nanoparticles of the protein. Prepn. of nanoparticles e.g. of human serum albumin, of elastin, and of human serum albumin with bound doxorubicin or Na salicylate is described, as are nanoparticle prepn. in the presence of salt and ue of various solvent systems in nanoparticle prepn.
