Abstract

Different polymers like Et cellulose, HP-55, HPMC K4M, were evaluated as microencapsulating agent and to prep. gastroretentive floating microspheres. Cefpodoxime proxetil is a third generation cephalosporin drug was selected as novel drug for the expt. The floating microspheres of Cefpodoxime proxetil were prepd. by non-aq. solvent evapn. techniques using different polymers as a rate controlling polymer and the microspheres were evaluated. The polymer coated microspheres are spherical, smooth, free flowing type. Microencapsulation efficiency was in the range between 72.41-88.96 %. The FTIR and DSC study confirmed that there is no chem. interaction during encapsulation process. The effect of polymer on release profile of drug was calcd. Cefpodoxime proxetil release from the polymer coated microspheres was slow over 12 h and dependant on core: coat ratio, wall thickness and size of microspheres. Drug release was by non-fickian diffusion mechanism. Good linear relationships were obsd. between wall thickness of the microspheres and release rate. Polymer coated microspheres of Cefpodoxime proxetil exhibited good prolong release characteristics and were found suitable for once a day oral controlled release products. [on SciFinder(R)]
