Abstract

Introduction: This study aimed to improve our gas chromatography/mass spectrometry screening method by using a deconvolution software (AMDIS) and a SIM/SCAN mode of detection. Methods: Gas chromatography-mass spectrometry analyses were carried out on an Agilent Technologies equipment. Data acquisition and analysis were performed using different softwares: Retention Time Locking, AMDIS and MSD Chemstation, as well as 2 commercial libraries and an in-house library. Two plasmas were prepared: 16 drugs of interest were added to the plasma A so as to test AMDIS, whereas plasma B was prepared by adding buprenorphine and norbuprenorphine to a final concentration of 1 $\mu$g/L to control the sensitivity of the SIM/SCAN method. Results: The analyse of plasma A shows that 11 drugs were detected by using AMDIS whereas only 5 were identified with the commercial libraries. This GC-MS method does not allow the identification 5 drugs among these 16 tested drugs. It can be noticed that buprenorphine and norbuprenorphine can be detected only by using SIM/SCAN acquisition mode. Conclusion: This deconvolution software allows to distinguish the spectrum of the compound from the background noise and the co-elutants. Furthermore, with the SIM/SCAN acquisition mode, the number of injections was able to be decreased.
