Abstract
Introduction: Leprosy is still a health problem in Indonesia. Vitamin D receptors are known to affect leprosy. Polymorphism of the VDR gene can lead to deregulation of vitamin D. The study aimed to evaluate the prevalence of polymorphism of the vitamin D receptor gene FokI rs2228570 in patients with multibacillary type leprosy. Methods: This study was a cross-sectional study with a purposive sampling method. Patients were enrolled on an outpatient basis between October - December 2023. A total of 27 patients were selected from inclusion and exclusion criteria. Gene amplification was done by using PCR and sequencing procedures. The data was analyzed using SPSS ver. 25, and a p-value ≤ 0.05 was considered significant. Results: Based on the results of gene sequencing, it was found that the genotype VDR FokI rs2228570 is most Ff, with 13 patients (48.14%), followed by ff and FF genotypes with 8 patients (29.62%) and 6 patients (22.22%). Conclusion: Based on research results, the prevalence of VDR FokI rs2228570 genotype polymorphism is the most commonly found variant. Pendahuluan: Kusta masih menjadi permasalahan kesehatan yang terjadi di Indonesia. Reseptor vitamin D diketahui berpengaruh terhadap kusta. Polimorfisme dari gen VDR dapat menyebabkan deregulasi dari vitamin D. Penelitian ini bertujuan untuk mengevaluasi prevalensi polimorfisme gen reseptor vitamin D FokI rs2228570 pada pasien kusta tipe multibasiler. Metode: Studi ini adalah studi dengan desain obervasional retrospektif dengan metode purposive sampling. Pasien merupakan pasien sudah sedang dalam terapi kusta yang dirawat di RSUP Prof. Dr. I.G.N.G. Ngoerah antara bulan Oktober - Desember 2023. Total 27 pasien dipilih berdasarkan kriteria inklusi dan eksklusi. Amplifikasi gen dilakukan menggunakan prosedur PCR dan sekuensing. Data dianalisis menggunakan SPSS ver. 25, dan nilai p ≤ 0,05 dianggap signifikan. Hasil: Berdasarkan hasil sekuensing gen, didapatkan bahwa genotipe gen VDR FokI rs2228570 terbanyak adalah genotipe Ff, yakni sebanyak 13 pasien (48,14%), diikuti dengan genotipe ff dan FF masing-masing sebanyak 8 pasien (29,62%) dan 6 pasien (22,22%). Simpulan: Berdasarkan hasil penelitian, prevalensi polimorfisme gen VDR FokI rs2228570 genotipe heterozigot Ff merupakan varian yang paling banyak ditemukan.
