Abstract

19randomamplifiedpolymorphicDNA(RAPD)andfiveInter-simplesequencerepeat(ISSR)primers wereusedtocharacterize35potatoaccessionsoriginatingfromMexico,EuropeandU.S.A,withthe aimofgeneratinginformationonthegeneticdiversityandrelationshipamongthegenotypesforbetter exploitationinbreedingprograms.Atotalof222and82lociweregeneratedbytheRAPDandISSR primers,respectively.BothRAPDandISSRprimersrevealed81.45and82.98%polymorphism, respectively.ManteltestshowednocorrelationbetweenRAPD andISSR (0.27),RAPD andRAPD +ISSR (0.47)similaritymatrices.However,ahighlevelofcorrelation(0.74)wasobservedbetweenISSRand RAPD+ISSRsimilaritymatrices.TheISSRmarkerwasfoundtobemoreefficientcomparedtoRAPD marker,therebyinfluencingmoretheconsensusdata.Theuseofconsensusdatageneratedmore informationrelatedtogeneticdiversityinpotato.RAPDandISSRmarkerssuccessfullydifferentiated betweenthepotatoaccessionsandsubgroupedthegenotypesbasedonorigin.Informationongenetic diversity andrelationshipswillbeusefulinthe selectionofparents andmappingstudies
