Abstract

Background: The apoE4 mutant apoE4[L28P] is associated with an increased risk for Alzheimer disease (AD) in addition to the known effect of apoE4. Results: ApoE4[L28P] displays folding defects and aberrant functions associated with AD pathogenesis. Conclusion: The structural integrity of apoE4 is an important component of its role in AD pathogenesis. Significance: We provide insights into the molecular basis for the added risk for AD in apoE4[L28P] carriers. © 2014 by The American Society for Biochemistry and Molecular Biology, Inc.
