Study of immobilization of nuclease P1 on paper cellulose

Abstract

In this study, immobilization of nuclease p1 on paper cellulose was studied. Effect of various factors, such as etherification pH, couple pH and time, (NH4)2SO4 concentration, enzyme amount et al, on the activity of immobilized enzyme was investigated. The immobilization conditions were optimized by orthogonal experiment. Characterizations of immobilized nuclease p1 were also evaluated. Through orthogonal experiment, the optimal immobilization conditions were as follows: couple pH 12, etherification pH 7.0, (NH4)2SO4 concentration 10% and enzyme amount 6.0 ml. Optimal pH of immobilized enzyme was 5.8. Optimal temperature of immobilized enzyme was 80°C. Thermal, operational and storage stabilities of the enzyme were improved after it was immobilized on paper cellulose. Michaelis constant Km of immobilized enzyme was disclosed as 5.72 g/l by the Lineweaver-Burk plot at 70°C.
