Abstract
Abstrak Teknologi diagnosis klinik yang murah dan andal merupakan sasaran pengembangan teknologi kesehatan untuk negera berkembang. Pada penelitian ini, teknik deteksi kandungan glukosa darah dikembangkan untuk diagnosis penyakit diabetes. Pemisahan plasma darah dilakukan dengan menggunakan peranti mikrofluida berbasis kertas yang terlapisi partikel nano ZnO bermorfolofi agregat sferis (SPs) dan nanoflowers (NFs), serta mekanisme pemisahan plasma darah dievaluasi secara numerik menggunakan modul aliran dalam media berpori pada perangkat lunak CFD (computational fluid dynamics). Kadar glukosa dalam plasma darah diukur dengan menggunakan spektroskopi FTIR (Fourier Transform Infrared) dan Raman. Hasil evaluasi peranti mikrofluida menunjukkan karakteristik watu pemisahan plasma darah berkisar dari 2 – 6 menit. Variasi waktu pemisahan plasma darah disebabkan topografi permukaan partikel nano ZnO yang tidak homogen pada kanal pemisahan. Pengukuran kadar glukosa berbasis spektroskopi FTIR dan Raman juga menunjukkan korelasi linier yang positif dan signifikan (R 2 > 0,9). Secara spesifik, monitoring nilai absorpsi IR pada frekuensi 1070 cm -1 dan hamburan Raman pada frekuensi 1121 cm -1 menghasilkan pengukuran dengan korelasi linier yang tinggi dan sensitivitas yang baik. Abstract A reliable and affordable clinical diagnostic technology constitutes a target of medical technology development in the developing countries. In this work, detection technique of blood glucose level is developed for diagnosis of diabetes. Blood plasma separations were carried out by using paper-based microfluidic devices decorated with ZnO nanoparticles whose morphology formed spherical aggregates (SPs) and nanoflowers (NFs). A detailed mechanism of blood plasma separation was evaluated by using an established module of flow over porous medium in computational fluid dynamics (CFD) software. Glucose concentration in the separated blood plasma were measured by using FTIR (Fourier Transform Infrared) and Raman spectroscopy. Evaluation of microfluidic devices reveals that the time required for blood plasma separation spans 2 – 6 mins. Different times required for blood plasma separation are possibly due to inhomogeneous surface topography of ZnO nanoparticles on the top of separation channel. Measurements of glucose concentration by means of FTIR and Raman spectroscopy reveal a significant linear correlation (R 2 > 0.9). In particular monitoring the IR absorption at 1070 cm -1 and Raman scattering at 1121 cm -1 results in high linear correlation and sensitivity.
