Abstract

Isolation and Morphological Identification and Phosphate Dissolution Test against Rhizosphere Bacteria in Soybean (Glycine max L.). Supervised by  Suriyanti, and Maimuna Nontji. This research was conducted to obtain bacterial isolates from the results of isolation and morphological identification of Rhizosphere bacteria in soybean plants, to obtain bacterial isolates capable of dissolving phosphates and calculating the clear zones of bacterial isolates. The research was carried out at the Microbiology Laboratory, Faculty of Pharmaceutical Sciences and at the Pest Laboratory and The Faculty of Agriculture, Muslim University of Indonesia will take place from December 2019 to February 2020. The materials to be used are heat-resistant plastic, aluminum foil, plastic wrap for soil samples of soybean Rhizosphere, nutrient agar (NA) media, phykovskaya media, yeast media. extract mannitol agar (YEMA), congo red (CR), bromine thymol blue (BTB) and 1000 ml distilled water.The results of isolation and identification of bacterial isolates from the rhizosphere of soybean plants (Glycine Max.L) were obtained as many as 23 isolates with various characteristics, almost all of which were milky white and soft in consistency by testing 8 isolates from 23 isolates with phycovkaya media showing 2 isolates were able to dissolve phosphate with an average Phosphate Solvent Index for isolate 13 of 1.5 and isolate 20 of 0.75. By testing on YEMA media with both Congo Red and Brom thymol blue indicators, no isolates were identified as Rhizobium bacteria due to contamination.
