Abstract

A review. An in vivo directed evolution strategy has been demonstrated to be a simple and efficient method to increase the thermostability of firefly luciferase, which allows one to obtain highly thermostable mutants without sacrificing catalytic efficiency,. A final mutant obtained in this way even displayed superior catalytic properties such as higher specific activity, lower Km for ATP, and increased temp. optimum. [on SciFinder(R)]
