Antioxidant and anti-inflammatory activities of bioactive peptides of gelatin extracted from Lates calcarifer scales using ultrasonication method

Abstract

Fish-derived peptides, particularly gelatin (G) and gelatin peptides (GP) that are extracted from Lates calcarifer scales through ultrasonication, have various biological activities. The gelatin peptide fraction (GPF) is produced by separating GP based on the differences in molecular weight. Therefore, this research aimed to determine the potential antioxidant and anti-inflammatory activities in vitro of GP and GPF derived from Lates calcarifer scales. The amino acid analysis of GPF showed the five most abundant amino acids, including alanine, tyrosine, phenylalanine, lysine, and cysteine. The antioxidant activities of GP and GPF, assessed through FRAP and ABTS methods, indicated IC50 values comparable to the positive standard. Specifically, GPF<3 showed superior antioxidant activities using the ABTS method with an IC50 value of 19.45±0.05 μg/mL, closely related to the Trolox value. Additionally, GPF<3 showed the best anti-inflammatory activities, with an IC50 value of 5.42±0.65 μg/mL, surpassing the efficacy of diclofenac. The novel GP derived from Lates calcarifer showed significant antioxidant and anti-inflammatory activities in vitro, suggesting potential for further investigations in animal research.
