Abstract
In this chapter, the rationale behind methods of design and analysis of genetic association studies will be reviewed and discussed, and the similarities between genetic association studies and classic epidemiological studies of environmental risk factors will be described. Issues that are specific to studies of genetic risk factors such as the use of certain family-based designs, accounting for different underlying genetic mechanisms and the impact of population history, will also be discussed.
