Abstract
The skin has a considerable protective role against harmful external factors, so the integrity of its structure is essential. Caffeine and ascorbyl magnesium phosphate are valuable substances in the treatment of skin lesions. Therefore, the aim of the current research was the preparation, physico-chemical characterization and pharmaco-toxicological evaluation of poloxamer-based hydrogel formulations with caffeine and magnesium ascorbyl phosphate (CAF-P407, MAP-P407 and CAF-MAP-P407) for topical application in the treatment of skin lesions. From a physico-chemical and rheological point of view, all experimental formulations showed suitable properties as in situ gel systems intended for skin application. In vitro, CAF-P407, MAP-P407 and CAF-MAP-P407 exhibited biocompatibility on healthy human HaCaT and JB6 Cl 41-5a skin cells. Moreover, the hydrogels revealed a wound-healing action by stimulating the migration of skin cells. In conclusion, the hydrogel formulations can be optimal for the topical administration of caffeine and ascorbic acid derivative in skin pathologies.
